Leveraging Synthetic DNA in AAV Manufacturing to Increase Efficiency
Transient transfection remains the most widely used method for AAV manufacturing, but it can be difficult to optimize due to competing process requirements. Factors such as cell density, DNA concentration, transfection reagent toxicity, and transfection mix characteristics must be carefully balanced to achieve high productivity and consistent performance. In addition, plasmid DNA represents a significant production cost and can introduce unwanted impurities, including bacterial backbone sequences that may be released during cell lysis or inadvertently packaged into AAV capsids.
This poster explores the potential of synthetic, enzymatically produced DNA as an alternative to conventional plasmid DNA for AAV production. By eliminating bacterial backbone sequences, synthetic DNA offers an attractive safety profile and may help address key manufacturing challenges. The study evaluates the performance of synthetic DNA compared with traditional plasmid DNA and assesses its suitability for scalable AAV manufacturing.
